hPSCs were damaged while they were harvested. Ideally hPSCs should be lifted only by pipetting several times using Solution D1 as instructed in the protocol. If pipetting requires more than 15 times, reduce the concentration of the substrate used to coat the dish/well for the maintenance of the source hPSCs to 70% or 50%.

We have observed that 4-well plates available from Nunc sometimes do not allow cells to seed in the periphery of the well. Next time, please try the following: 1. switch to 24-well plates; 2. incubate the plate with Coating Material A at 4°C overnight, instead of 37°C for 2 hours; 3. do not touch the bottom of wells with fingers when handling the plate.

All reagents in our kits are provided 10% more than needed. Please check if pipettors used are calibrated recently. On the other hand, particularly for the components and the coating materials, a small volume may have been on a tube wall. Breif centrifugation of those tubes will bring the whole content to the bottom. Based on our data, for example, the percentages of TUBB3+neurons differentiated using Quick-Neuron™ Cholinergic mRNA and SeV were 93% and 88%, respectively. Thus, there is not much differences in the differentiation efficiency between an mRNA-based and a SeV-based kit.

Unlike other human cells, hPSCs form disc-shaped colonies with tightly packed cells if their pluripotency is maintained. Thus, it is advisable to confirm the pluripotency of hPSCs by their appearance before use. It takes at least 3-4 days for colonies with the typical appearance to emerge after plating at 1,000 cells / cm2. When an hPSC culture reaches 80% confluency, the culture must be used or passaged.

Use a P20 pipettor and make a small drop of SeV solution on the pipet tip as the plunger is slowly pushed down. Since the drop can touch the culture medium, repeat making a drop and touching the medium severall times at different sites within the culture.

SeV is not pathogenic to humans (i.e., human is not the natural host of the virus) and the infection does not persist in immunocompetent animals. Furthermore, SeV used in our kits do not produce infectious viral particles upon transduction to host hPSCs and is a temperature-sensitive mutant, such that it is active at 33℃ but becomes inactive at 37℃. However, because SeV can be transmitted by aerosol and contact with respiratory secretions and is highly contagious, appropriate care must be taken to prevent the potential mucosal exposure to the virus. Collectively, our SeV-based kits must be used under Biosafety Level 2 (BL-2) containment with a biological safety cabinet or a laminar flow hood, and with appropriate personal protective equipment. In the event that the virus comes into contact with skin or eyes, decontaminate the affected area by flushing with a plenty of water and follow the safety manual prepared by your laboratoy that is approved by the Institutional Biosafety Committee.

These two culture conditions permit plating hPSCs at a lower density or provide a better cloning efficiency of single hPSCs, and have been producing a high quality differentiation using our mRNA-based kits. Before starting incubation, please make sure that cover slips are submerged in Coating Material B or C; they can easily be under the liquid by shaking and/or rocking the plate. Alternatively, the cover slips can be flipped since the other side must have been coated.

Please perform passaging on Day 3 as directed in the protocol becuase it is anticipated that the differentiation of neurons is less efficient due to the outgrowth of precursor cells. Passaging on Day 3 will result in enriching neurons in newly prepared cultures. In addition, if you plan to use our kits again, please perform the following test: count and plate 0.1, 0.2, 0.5 and 1.0 x 10^5 cells per well and determine the best cell count that results in a similar cell denstiy to the one shown as the Day 1 image in the protocol.

We have tested and confirmed the expression of the following markers: choline acetyltransferase (ChAT, a cholinergic neuron marker), tyrosine hydroxylase (TH, a dopaminergic neuron marker), parvalbumin (PVALB, a GABAergic neuron marker), vesicular glutamate transporter 1 (vGLUT1, a glutamatergic neuron marker), tryptophan hydroxylase 2 (TPH2, a serotonergic neuron marker).
